The Undead Jester spawns at the blue dot and walks along the green line shown on the map below.  To spawn him, you need to kill every mob that walks along that green line.  You don't have to kill any other walker - some walk partially along the green line and turn back or go into one of the rooms.  His placeholder walks the full length of the green line.  All you have to do is sit at the location marked by the star and look south.
